International mobility for PhD
Nicole Vercellino is working on the study of the distribution and role of senescent macrophages in the synovial membrane of osteoarthritis patients (University of Piemonte Orientale).
Research project: Senescent cells accumulate in joint tissues in response to injury and aging, and may contribute to the development and perpetuation of osteoarthritis (OA). In particular, synovial immunosenesence has largerly been unexplored in the context of OA. Macrophages are the main immune cells present in synovial tissue, they undergo dysregulations with aging and can become senescent themselves and/or participate in the propagation or accumulation of cellular senescence in the joint. This project aims to better characterize senescent macrophages, their distribution, their role in OA synovium as well as their interplay with surrounding joint cells. A validated senescent cells geneset will be applied to scRNA-seq data performed on synovial tissues presenting distinct pathotypes. Moreover, multiple immunofluorescence staining will be performed to evaluate the senescence markers expression in synovial macrophages in OA synovial tissues. Co-culture experiments will also be conducted to identify how senescent macrophages can affect joint cells and the mediators involved.
